研究了偶氮胂K(Arsenazo K) 与蛋白质的反应及利用此反应测定蛋白质的最佳实验条件。 偶氮胂K 与蛋白质生成的复合物的最大吸收波长为611 nm , 比偶氮胂K 红移71 nm , 对照性好。 在室温下, 于pH 2.8 ~3 .9的缓冲溶液中, 反应迅速完成。 表面活性剂Triton X- 100 和溴化十六烷基吡啶(CPB) 均可提高反应的灵敏度,复合物稳定, 干扰少。 绘制了牛血清白蛋白(BSA) 、γ- 球蛋白( γ_G) 、人血清白蛋白( HAS) 、血红蛋白(Hb) 、卵白蛋白( OVA) 、溶菌酶(Lyso) 、胃蛋白酶(Pep) 、糜蛋白酶(Chy) 的工作曲线。测定了人血清中的蛋白质总量,结果可靠。
The reaction between arsenazo K (Arsenazo K) and protein and the optimum conditions for the determination of protein using this reaction were studied. The maximum absorption wavelength of the complex of arsenazo K and protein was 611 nm, which was 71 nm better than that of arsenazo K red dye. At room temperature, at pH 2.8 ~ 3. 9 buffer solution, the reaction is completed quickly. Surfactants Triton X-100 and cetyl pyridinium bromide (CPB) can improve the sensitivity of the reaction, the stability of the complex, less interference. Bovine serum albumin (BSA), γ-globulin (γ_G), human serum albumin (HAS), hemoglobin (Hb), ovalbumin (OVA), lysozyme (Lyso), Pep, Protease (Chy) working curve. The total amount of protein in human serum was measured and the result was reliable.
