目的通过研究千佛菌对D-半乳糖致衰老模型小鼠外周血Th1、Th2型细胞因子(IFN-γ、IL-10)水平及IFN-γ/IL-10比值的影响,探讨千佛菌对衰老机体的免疫调节作用。方法 将小鼠随机分为正常对照组、衰老模型组、千佛菌大剂量组、千佛菌小剂量组。除正常组外,其余均腹腔注射D-半乳糖制备衰老模型。千佛菌大、小剂量组分别以100%和50%药液0.4 mL/只/日灌胃,其余等量生理盐水灌胃。测定血清中IFN-γ、IL-10含量并计算IFN-γ/IL-10比值,测定脾脏及胸腺指数。结果 模型组小鼠血清IFN-γ、IFN-γ/IL-10比值、脾脏及胸腺指数均明显降低,IL-10水平显著升高。与正常组比较,差异显著(P<0.001、P<0.05);与模型组比较,千佛菌大、小剂量组IFN-γ、脾脏及胸腺指数均明显升高,而IL-10水平明显降低。差异显著(P<0.001,P<0.05),IFNγ/IL-10比值恢复;千佛菌大、小剂量组间IFN-γ、IL-10比较无差异(P>0.05)。结论 千佛菌对D-半乳糖致衰老模型小鼠Th1、型细胞因子IFN-γ水平及IFN-γ/IL-10比值具有升高作用,可使血清中Th2型细胞因子IL-10降低,并能够显著增加小鼠的脾脏及胸腺指数,对D-半乳糖导致衰老的Th1/Th2免疫失衡具有一定的调节作用,能使之趋向恢复正常。
Objective To investigate the effects of Thousand Flavus on the levels of Th1 and Th2 cytokines (IFN-γ, IL-10) and the ratio of IFN-γ / IL-10 in peripheral blood of D-galactose- Immune regulation of the aging body. Methods The mice were randomly divided into normal control group, aging model group, high-dose group of Thousand Flaviviridae, low-dose group of Thousand Flaviviridae. In addition to the normal group, the rest were intraperitoneal injection of D-galactose preparation aging model. Thousands of bacteria large and small dose group were 100% and 50% liquid 0.4 mL / only / day gavage, and the remaining equal amount of saline gavage. Serum levels of IFN-γ and IL-10 were measured and the ratio of IFN-γ / IL-10 was calculated. The indexes of spleen and thymus were measured. Results The serum levels of IFN-γ, IFN-γ / IL-10, spleen and thymus index in model group were significantly decreased and IL-10 levels were significantly increased. Compared with the normal group, the difference was significant (P <0.001, P <0.05). Compared with the model group, IFN-γ, spleen and thymus index of the large and small doses of the group of Thunbergii were significantly increased, while the level of IL-10 was significantly decreased . (P <0.001, P <0.05), and the ratio of IFNγ / IL-10 recovered. There was no significant difference in the levels of IFN-γ and IL-10 between the groups of large and small doses of Michelia philoxeroides (P> 0.05). CONCLUSION: Thielaviae can up-regulate Th1 cytokine IFN-γ and IFN-γ / IL-10 ratio in D-galactose-induced aging mice and decrease Th2 cytokine IL-10 in serum. And can significantly increase the mouse spleen and thymus index, D-galactose-induced aging Th1 / Th2 immune imbalance has a certain regulatory role, can make the trend back to normal.
