目的探讨Stathmin/oncoprotein18(Op18)基因在人脑胶质瘤中的表达规律及意义。方法分别采用免疫组化法和Western blot法检测10例正常脑组织和56例脑胶质瘤中Stathmin蛋白的表达。结果免疫组化法检测Stathmin蛋白在正常脑组织、低级别胶质瘤(Ⅰ~Ⅱ级)及高级别胶质瘤(Ⅲ~Ⅳ级)中阳性表达率分别为20%、65%、100%。正常脑组织分别与Ⅰ~Ⅱ级组、Ⅲ~Ⅳ级组比较,均有显著性差异(P<0.05);Ⅰ~Ⅱ级组与Ⅲ~Ⅳ级组比较,有显著性差异(P<0.05)。Western blot法检测显示,Stathmin蛋白在胶质瘤中的表达明显增高,正常脑组织分别与Ⅰ~Ⅱ级组、Ⅲ~Ⅳ级组比较,差异均有显著性(P<0.01),Ⅰ~Ⅱ级组与Ⅲ~Ⅳ级组比较,差异有显著性(P<0.01)。结论Stathmin在脑胶质瘤中过表达,Stathmin可能为脑胶质瘤的生物治疗提供一个新靶点。
Objective To investigate the expression and significance of Stathmin / oncoprotein18 (Op18) gene in human glioma. Methods The expressions of Stathmin protein in 10 normal brain tissues and 56 gliomas were detected by immunohistochemistry and Western blot respectively. Results The positive rates of Stathmin protein in normal brain tissue, low grade gliomas (grade Ⅰ ~ Ⅱ) and high grade gliomas (Ⅲ ~ Ⅳ grade) were 20%, 65% and 100% . There were significant differences between normal group and Ⅰ ~ Ⅱ group and Ⅲ ~ Ⅳ group (P <0.05). There was significant difference between Ⅰ ~ Ⅱ group and Ⅲ ~ Ⅳ group (P <0.05) ). The results of Western blot showed that the expression of Stathmin protein in gliomas was significantly higher than that in normal brain tissues (P <0.01), Ⅰ ~ Ⅱ There was significant difference between grade group and grade Ⅲ ~ Ⅳ group (P <0.01). Conclusion Stathmin is overexpressed in gliomas, Stathmin may provide a new target for the biological treatment of glioma.
