目的探讨川芎嗪(TMP)对大鼠脊髓急性损伤(ASCI)模型脊髓神经生长因子(NGF)、脑源性神经生长因子(BDNF)表达的影响及其对脊髓损伤后脊髓功能的保护作用。方法取SD大鼠随机分为正常对照组、假治疗组和川芎嗪组并给予相应处理。采用改良Allen’s法造模,BBB评分对实验大鼠脊髓功能进行行为学评分;SABC法检测造模后大鼠损伤段脊髓NGF和BDNF的表达。结果随时间延长,术后SD大鼠BBB评分逐渐升高,术后7 d、14 d和21 d川芎嗪组BBB评分值均较假治疗组高(P均<0.05);川芎嗪组术后3 d7、d和14 d时NGF、BDNF阳性细胞数表达均较假治疗组明显增加(P均<0.05)。结论川芎嗪对脊髓继发性损伤有保护作用,这种保护作用可能与其能促进损伤段脊髓NGF、BDNF的表达有关。
Objective To investigate the effect of tetramethylpyrazine (TMP) on the expression of nerve growth factor (NGF) and brain-derived nerve growth factor (BDNF) in the spinal cord of rats with acute spinal cord injury (ASCI) and its protective effect on spinal cord function after spinal cord injury. Methods SD rats were randomly divided into normal control group, sham-treated group and ligustrazine group and given corresponding treatment. The modified Allen’s method and BBB score were used to evaluate the spinal cord function in experimental rats. The expression of NGF and BDNF in injured spinal cord of rats was detected by SABC method. Results With the extension of time, the score of BBB in SD rats gradually increased after operation, and the BBB scores of ligustrazine group at 7 d, 14 d and 21 d after operation were significantly higher than those in sham-treated group (all P <0.05) The expression of NGF and BDNF positive cells on 3 d7, d and 14 d were significantly higher than those in sham treated group (all P <0.05). Conclusion Tetramethylpyrazine has a protective effect on secondary spinal cord injury, which may be related to the promotion of NGF and BDNF expression in injured spinal cord.
