本研究在已构建人睾丸cDNA表达文库,并用抗人精子抗体从中筛选出人精子抗原基因表达克隆(HSG)的基础上,对其中HSG3克隆进行了生物学效应的测定,结果显示①HSG3克隆的溶原蛋白分子量为149KDa,并被兔抗人精子抗体识别。②HSG3克隆表达抗原的特异抗体(HSG3Ab)对人精子SIT和GAT无明显影响,但能阻断人精子在顶体反应时,顶体后区ConA受体的暴露。表明HSG3克隆的表达抗原具有受精抗原的特性,能作为精子免疫避孕疫苗的候选成份。
In this study, human testes cDNA expression library has been constructed, and the human sperm antigen gene expression clone (HSG) was screened by using anti-human sperm antibody, and the biological effect of HSG3 clone was determined. The results showed that ① HSG3 clone The original protein has a molecular weight of 149 kDa and is recognized by rabbit anti-human sperm antibodies. (2) HSG3Ab showed no significant effect on SIT and GAT of human spermatozoa, but blocked HSC3Ab exposure to acrosome reaction in the acrosome reaction. HSG3 clone expression antigen that has the characteristics of fertilized antigen can be used as a candidate component of sperm immunization contraceptive vaccine.
