通过抑制率及抑制动力学的测定,研究了红旱莲总黄酮(HAF)对α-葡萄糖苷酶的抑制作用。利用对1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除能力、β-胡萝卜素漂白抑制能力和抗DNA氧化损伤能力的测定,评估了红旱莲总黄酮体外抗氧化能力。红旱莲总黄酮是一种可逆的混合型α-葡萄糖苷酶抑制剂,其半抑制质量浓度(IC50)为0.237 mg/mL,抑制常数(K i)为0.149 mg/mL,酶–底物络合物抑制常数(K is)为0.123 mg/mL。红旱莲总黄酮有较强的DPPH自由基清除能力(IC50=2.71μg/mL),能够显著地抑制β-胡萝卜素的漂白及2,2’-偶氮二异丁基脒(AAPH)引起的DNA氧化损伤。红旱莲黄酮可作为潜在的抗氧化剂和预防治疗Ⅱ型糖尿病的药物或膳食功能因子进行深入研究与开发。
