目的 :探讨原发性肺癌组织中端粒酶反转录酶 (h TRT)的表达和端粒酶活性的关系以及在肺癌分子生物学诊断的临床意义。方法 :用免疫组化和 TRAP法分别观察 5 4例原发性肺癌和 10例正常肺组织 ,比较分析它们的相关性。结果 :h TRT阳性率 83% (4 5 / 5 4) ,端粒酶活性阳性率 85 % (4 6 / 5 4) ,在肺癌组中 3例术前化疗患者h TRT表达 2例呈弱阳性 1例阴性 ,肺癌淋巴结转移的 h TRT阳性率达 92 % ,5例小细胞肺癌 h TRT均为阳性 ,10例正常肺组织均为阴性。结论 :抗 h TRT抗体在肺癌中具有癌细胞的特征性 ,并与端粒酶活性呈正相关 ,可作为肺癌诊断的灵敏性和特异性的指标 ,用免疫组化法比 TRAP法检测肺癌中端粒酶的活性将更为简便易行
Objective: To investigate the relationship between telomerase reverse transcriptase (h TRT) expression and telomerase activity in primary lung cancer and its clinical significance in molecular diagnosis of lung cancer. Methods: Fifty-four cases of primary lung cancer and 10 cases of normal lung tissue were observed by immunohistochemistry and TRAP respectively. Their correlations were analyzed. Results: The positive rate of h TRT was 83% (45/54) and the positive rate of telomerase activity was 85% (46/54). In the lung cancer group, the expression of h TRT in 3 cases of preoperative chemotherapy was weakly positive One case was negative, the positive rate of hTRT in lymph node metastasis of lung cancer was 92%, h TRT in 5 cases of small cell lung cancer was positive, and 10 cases of normal lung tissue were negative. Conclusion: Anti-h TRT antibody has characteristic of cancer cells in lung cancer and is positively correlated with telomerase activity, which can be used as an indicator of sensitivity and specificity of lung cancer diagnosis. Compared with TRAP method, Granzyme activity will be more simple and easy
