目的探讨NF-κB在大肠癌细胞MMP-3蛋白诱导表达中的作用及抗氧化剂对大肠癌细胞MMP-3蛋白诱导表达的影响及其机制。方法培养上清MMP-3蛋白含量用ELISA检测,EMSA法测定细胞核内NF-κB结合活性。结果抗氧化剂可抑制大肠癌细胞培养体系TNF-α诱导的MMP-3蛋白生成,TNF-α可诱导大肠癌细胞NF-κB的激活并被抗氧化剂阻断。结论TNF-α诱导的大肠癌细胞MMP-3蛋白表达依赖于NF-κB的激活;抗氧化剂可通过抑制NF-κB的活化而阻断MMP-3蛋白的诱导表达。
Objective To investigate the role of NF-κB in the induction of MMP-3 protein in colorectal cancer cells and the effect of antioxidants on the induction of MMP-3 protein in colorectal cancer cells. Methods The content of MMP-3 in the culture supernatant was detected by ELISA, and the activity of NF-κB in the nucleus was detected by EMSA. Results Antioxidants could inhibit TNF-α-induced MMP-3 protein production in colorectal cancer cell culture system. TNF-αinduced the activation of NF-κB in colorectal cancer cells and was blocked by antioxidants. Conclusion TNF-αinduced the expression of MMP-3 protein in colorectal cancer cells, which was dependent on the activation of NF-κB. Antioxidants could block the induction of MMP-3 protein by inhibiting the activation of NF-κB.
