目的探讨肾细胞癌(RCC)组织中基质金属蛋白酶-2(MMP-2)与血管内皮生长因子(VEGF)表达与血管生成的关系。方法用免疫组织化学SP法检测57例肾细胞癌、9例正常肾组织中的MMP-2与VEGF的表达,计数微血管密度(MVD),血管生成采用CD34免疫组化染色方法标记微血管内皮。结果 RCC组织中MMP-2与VEGF表达明显高于正常肾组织(P<0.05),不同临床分期的RCC组织中MMP-2与VEGF表达差异有统计学意义(P<0.05),不同病理类型的RCC组织中MMP-2与VEGF表达差异无统计学意义。MVD与VEGF、MMP-2蛋白表达之间呈显著相关性(P<0.05);VEGF与MMP-2蛋白表达之间有显著相关性(P<0.05)。结论 MMP-2与VEGF的表达可反映RCC侵袭和转移潜能的生物学指标,对临床判断肾癌预后有着直接意义。
Objective To investigate the relationship between the expression of matrix metalloproteinase-2 (MMP-2) and vascular endothelial growth factor (VEGF) and angiogenesis in renal cell carcinoma (RCC). Methods The expressions of MMP-2 and VEGF in 57 cases of renal cell carcinoma and 9 cases of normal renal tissue were detected by immunohistochemical SP method. The microvessel density (MVD) was counted and angiogenesis was counted. Microvascular endothelium was labeled by immunohistochemical staining. Results The expression of MMP-2 and VEGF in RCC tissues was significantly higher than that in normal renal tissues (P <0.05). There was a significant difference in the expression of MMP-2 and VEGF in different clinical stages of RCC (P <0.05) The expression of MMP-2 and VEGF in RCC tissues showed no significant difference. There was a significant correlation between MVD and VEGF, MMP-2 protein expression (P <0.05). There was a significant correlation between VEGF and MMP-2 protein expression (P <0.05). Conclusions The expression of MMP-2 and VEGF may reflect the biological indexes of invasion and metastasis potential of RCC, which has a direct meaning for judging the prognosis of RCC.
