目的:探讨VEGF-B基因启动子在鼻息肉中甲基化状态。方法:应用MS-PCR检测28例实验组鼻息肉组织和12例对照组正常组织VEGF-B基因启动子甲基化状态,并对MS-PCR产物进行测序。结果:VEGFB启动子甲基化在28例鼻息肉中有9例阳性,19例阴性;在12例对照组织中8例阳性,4例阴性。鼻息肉组织和对照组织中VEGF-B基因启动子甲基化阳性率的差异有统计学意义(χ~2=4.096,P<0.05)。MS-PCR产物测序结果表明鼻息肉中存在VEGF-B基因启动子甲基化修饰。结论:鼻息肉中VEGF-B基因启动子可能存在低甲基化状态。推测鼻息肉VEGF-B基因甲基化调控着VEGF-B的表达,在鼻息肉的发生机制中占有重要地位。
Objective: To investigate the methylation status of VEGF-B gene promoter in nasal polyps. Methods: The methylation status of VEGF-B gene promoter was detected by MS-PCR in 28 cases of nasal polyps and 12 cases of normal controls. MS-PCR products were sequenced. Results: The methylation of VEGFB promoter was positive in 28 cases of nasal polyps, 19 cases were negative, 8 cases were positive in 12 cases and 4 cases were negative. The positive rate of VEGF-B promoter methylation in nasal polyps and control tissues was statistically significant (χ ~ 2 = 4.096, P <0.05). Sequencing results of MS-PCR products indicated that VEGF-B gene promoter methylation modification existed in nasal polyps. Conclusion: There may be hypomethylation of VEGF-B gene promoter in nasal polyps. It is speculated that the methylation of VEGF-B gene in nasal polyps controls the expression of VEGF-B, which plays an important role in the pathogenesis of nasal polyps.
