By light microscopy of stained mitotic chromosomes, one can see many of the structural abnormalities in the human Y chromosome. From the correlation between the phenotypes and the abnormal karyotypes it has been deduced that the determinants of the testis are located on the human Y chromosome. However, due to the limitations of the accuracy of chromosome band studies, the study of aberrant Y chromosome structure often shows inconsistent results. Therefore, it remains to be argued that the determinant of the testicle should be located on the short arm or centromere of the Y chromosome or on the long arm. Cloning of the Y-chromosome DNA sequence becomes a new tool for Y-chromosome analysis. It is now possible to identify some types of Y-chromosome DNA sequences. Y chromosome specific repeat
