目的:探讨五指毛桃对60Coγ射线损伤小鼠组织DNA的防护和修复作用。方法:120只SPF级雄性小鼠随机分为辐照前用药、辐照后用药、辐照前后连续用药三大模块各5组,各模块设低、中、高剂量给药组分别给予5、10、20 g/(kg·d)五指毛桃水提液连续灌胃7 d,正常对照组给予生理盐水,单纯照射组作为辐照模型组,除正常对照组外,所有小鼠接受全身一次性60Coγ照射,照射剂量为6Gy。各模块均于实验结束次日处死小鼠,取肺、肝、睾丸、股骨和外周血,分别制作单细胞悬液,用单细胞凝胶电泳技术进行DNA损伤检测。结果:辐照模型组小鼠组织细胞DNA的尾部DNA百分率(Tail DNA%)和尾矩(Tail Moment)显著高于正常对照组(P<0.05),各给药组的TailDNA%和Tail Moment均显著低于辐照模型组(P<0.01),且随着给药剂量的增加,Tail DNA%和Tail Moment降低。结论:五指毛桃水提液对60Coγ射线损伤小鼠组织DNA具有很好的防护作用和修复作用。
Objective: To investigate the protective and repair effects of peaches on DNA of 60Co γ-ray-injured mice. Methods: One hundred and twenty SPF male mice were randomly divided into pre-irradiation, post-irradiation and continuous administration of three groups of five groups before and after irradiation. Each group was given low, medium and high doses of 5, 10, 20 g / (kg · d) five finger peach water extract continuous gavage 7 d, the normal control group given saline, irradiation group as the irradiation model group, except for the normal control group, all mice received systemic one-time 60Coγ irradiation, irradiation dose of 6Gy. The mice were sacrificed on the following day after the experiment was completed. Lung, liver, testis, femur and peripheral blood were collected. Single cell suspensions were prepared, and DNA damage was detected by single cell gel electrophoresis. Results: Tail DNA% and Tail Moment of DNA in the irradiated model group were significantly higher than those in the normal control group (P <0.05). Tail DNA% and Tail Moment (P <0.01), and Tail DNA% and Tail Moment decreased with the increase of dose. Conclusion: The aqueous extract of Peach Blossom has good protective effect and repair effect on tissue DNA damaged by 60Co γ-ray.
