临床静脉注射IL-2时会产生明显的毒副作用,口服IL-2作为一种替代途径有望解决这一难题,但目前尚不清楚口服IL-2是否具有全身效应.为此作者通过给小鼠口服IL-2来检测其对外周血白细胞数和骨髓增殖的影响,来判断其是否产生全身效应.以6~8周龄G57BL/6小鼠作为实验动物,实验组分为四组,分别口服经明胶稀释的不同浓度的IL-2(40,400,4000,40000U/d);阳性对照组皮下注射经BSA和PBS稀释的IL-2(5000U/d),阴性对照组口服明胶稀释液或皮下注射BSA和PBS稀释液,连续给药三天后检测结果表明,口服IL-2(40,400,4000,40000U/d)的实验组,其外周血白细胞数分别降至未给药前加93.7±1.3%,83.6±2.6%,75.0±1.8%;75.4±2.3%;阳性对照组降至实验前的71.7±1.8%,而阴性对照组无明显差异.这表明小鼠外周血白细胞数随IL-2口服剂量的增加而降低.由于外周血白细胞数受骨髓功能的影响,为此进一步检测了小鼠口服IL-2后的骨髓功能.实验组口服IL-2(4000U/d)或皮下注射IL-2(5000U/d),阴性组口服明胶稀释液或皮下注射BSA和PBS稀释液,连续给药三天后杀死小鼠,取出骨髓细胞在含不同浓度M-CSF的培养基中培养七天,计算GM克隆数,测得在同一浓度加M-CSF中实验组得到的GM克隆数要少于对照组,说明口服的IL-2是通过抑制骨髓细胞的增殖来降低外
