目的:优化建立壁虎药材蛋白质的提取工艺。方法:通过比较超声和回流两种提取方式对壁虎药材蛋白质提取率的影响,筛选适合的提取方式。分别对pH、料液比、提取温度、提取时间4种影响因素进行单因素考察,并采用L9(34)正交试验设计,以蛋白提取率为指标,优选建立壁虎药材蛋白质的提取工艺。结果:超声提取的蛋白质含量显著高于回流提取,建立了壁虎药材蛋白质的最佳提取工艺:pH12,料液比1∶20,提取温度50℃,超声提取时间80 min。结论:该研究建立的蛋白质提取工艺稳定、合理、可行,为壁虎药材的质量评价和生物活性物质的进一步研究提供了实验依据。
Objective: To optimize the extraction of protein from gecko medicinal materials. Methods: By comparing the effects of ultrasound and reflux on the protein extraction rate of Gekko gecko, the suitable extraction methods were screened. The influence factors of pH, solid-liquid ratio, extraction temperature and extraction time were investigated by single factor method. The L9 (34) orthogonal design was used to determine the extraction rate of protein from gecko medicinal plants. Results: The protein content of ultrasonic extraction was significantly higher than that of reflux extraction. The optimal extraction process of protein was established: pH12, ratio of solid to liquid 1:20, extraction temperature 50 ℃, ultrasonic extraction time 80 min. Conclusion: The protein extraction process established in this study is stable, reasonable and feasible, which provides experimental evidence for the quality evaluation of gecko medicinal materials and further research on bioactive substances.
