目的 探讨湖南汉族人群 HL A- DP、HL A- DQ位点等位基因多态性与鼻咽癌遗传易感性之间的关系。方法 应用聚合酶链反应 -特异性寡核苷酸探针基因分型技术对 87例湖南汉族鼻咽癌患者与91名健康对照作 HL A- DPA1、- DPB1、- DQA1及 - DQB1的基因分型 ,采用 χ2检验比较两组各位点等位基因频率 ,单倍型频率分布的差异。结果 发现鼻咽癌组 DPA1* 0 2 0 1、DPB1* 190 1、DQA1* 0 2 0 1较对照组明显降低 ,DPB1* 0 4 0 2、DQA1* 0 10 1较对照组明显升高 ;单倍型 DPA1* 0 2 0 1- DPB1* 14 0 1及 DQA1*0 2 0 1- DQB1* 0 2 0 1较对照明显降低 ;但 P值经 Bonferroni校正后 ,差异均无显著性 ( Pc>0 .0 5 )。结论 湖南汉族人群 HL A- DP和 - DQ位点与鼻咽癌无明显相关 ,与以往用受累同胞对方法在鼻咽癌家系中未能证实鼻咽癌易感基因与 HL A- DP和 - DQ位点连锁的结果一致
Objective To investigate the relationship between HL-DP and HLA-DQ locus alleles and genetic susceptibility to nasopharyngeal carcinoma in Hunan Han population. Methods The genotypes of HL-DPA1, - DPB1, - DQA1 and - DQB1 in 87 Han nationality patients with NPC and 91 healthy controls in Hunan were genotyped by polymerase chain reaction-specific oligonucleotide probe. Type, usingχ2 test to compare the two groups of alleles frequency, haplotype frequency distribution differences. The results showed that DPA1 * 0 2 0 1, DPB1 * 190 1, DQA1 * 0 2 0 1 in nasopharyngeal carcinoma group were significantly lower than those in control group, DPB1 * 0 0402 and DQA1 * 0 10 1 were significantly higher than those in control group; DPA1 * 0 2 0 1-DPB1 * 14 0 1 and DQA1 * 0 2 0 1-DQB1 * 0 2 0 1 were significantly lower than those of the control; however, there was no significant difference in P value after Bonferroni calibration (Pc> 0 .0 5). Conclusion There is no significant correlation between HL-DP and - DQ loci and NPC in Hunan Han population. Compared with the previous study, DQ linkage of the results of the same site
