目的 检测胃癌患者 19号染色体微卫星位点的杂合性缺失 (less of heterozygosity,L OH) ,以初步确定 19号染色体上与胃癌相关基因连锁最密切的微卫星多态位点。方法 应用多重 PCR对 44例原发性胃癌患者中覆盖 19号染色体上的 2 2个微卫星位点 (遗传距离在 1.1~ 10 .9c M之间 )进行扩增。聚丙烯酰胺凝胶电泳分离 PCR产物 ,Gene Scan TM,Genotype TM软件进行分析。结果 19个位点 (19/2 2 )检测出 L OH阳性 ,总 L OH频率为 5 9% (2 6 /4 4) ,其中 D19S5 71位点的 L OH频率最高 (2 1.43% )。结论 高频的 L OH位点附近 ,可能存在未知的胃癌相关基因
Objective To detect the lack of heterozygosity (L OH) in the chromosome 19 microsatellite locus of gastric cancer patients to identify the microsatellite polymorphism sites closely linked with gastric cancer-related genes on chromosome 19. Methods Multiplex PCR was used to amplify 22 microsatellite loci on chromosome 19 (genetic distance between 1.1 and 10 .9 cM) in 44 patients with primary gastric cancer. The PCR products were separated by polyacrylamide gel electrophoresis and analyzed by Gene Scan TM and Genotype TM software. Results Positive LOH was detected at 19 sites (19/2 2 ). The total L OH frequency was 5 9% (2 6 /4 4). The DOH frequency of D19S5 71 site was the highest (2 1.43%). Conclusion There may be an unknown gastric cancer-related gene near the high-frequency L OH site
