本文报道用反相高效液相色谱法测定商品制剂中胰岛素的新改良法。过去报道各法均因Rr相似或制剂中防腐剂的干扰而受限制。改良法不受该因素干扰,测试结果与标示效价相符。材料和溶液:牛和猪的单组分胰岛素来自Novo工厂,生物鉴定标示效价:牛胰岛素27.2IU/mg、猪胰岛素26.8IU/mg、脱酰胺基胰岛素系由胰岛素在0.01N盐酸中、37℃
This article reports a new improved method for the determination of insulin in commercial preparations by reversed-phase high performance liquid chromatography. It has been reported in the past that each method is limited by the similarity of Rr or the interference of preservatives in the formulation. The modified method is not affected by this factor, the test results consistent with the labeling potency. Materials and Solutions: The bovine and porcine single-component insulin was from Novo Biotech. Bioassay labeling potency: bovine insulin 27.2 IU / mg, porcine insulin 26.8 IU / mg, deamidated insulin from insulin in 0.01 N hydrochloric acid, 37 ℃
