运用Dexter体外细胞长期培养法,对脐带血和外周血造血干细胞进行单独培养,用MRC-5细胞株作为基底层与之混合培养,将二者进行对照研究。结果发现外周血和脐带血不含或仅含极少数基质细胞,单独培养时造血干细胞的增殖分化受到影响;而用MRC-5细胞株作为基底层的混合培养中,CD+34可以在5周~6周内持续存活,并得到增殖分化,揭示MRC-5细胞株具有基质细胞的支持作用,为脐带血和外周血CD+34在体外的长期培养提供了简便可行的技术方法。
The cord blood and peripheral blood hematopoietic stem cells were cultured in vitro using the Dexter in vitro cell culture method. The MRC-5 cell line was used as the basal layer to mix and culture the two cells. As a result, it was found that peripheral blood and umbilical cord blood contained no or only a few stromal cells, and the proliferation and differentiation of hematopoietic stem cells when cultured alone was affected; however, in a mixed culture using MRC-5 cell line as a basal layer, CD+34 can be maintained for 5 weeks to 6 weeks. Within weeks of continuous survival and proliferation and differentiation, revealed that MRC-5 cell line has the supportive role of stromal cells, providing a simple and feasible technical method for long-term culture of cord blood and peripheral blood CD+34 in vitro.
