为了方便研究人脐静脉内皮细胞在造血调控和心血管系统中的作用 ,利用脂质体介导基因转染的方法 ,将SV4 0大T抗原导入原代人脐静脉内皮细胞 ,建立了至少可传代培养 2 5代的人脐静脉内皮细胞系IEC。免疫细胞化学染色显示IECⅧ因子阳性。流式细胞术检测I型荆豆凝集素结合率为 97%。染色体分析表明IEC细胞内染色体结构和数目发生了改变。体外实验表明 ,IEC具有支持造血的能力
In order to facilitate the study of the role of human umbilical vein endothelial cells in hematopoietic regulation and cardiovascular system, SV40 T antigen was introduced into primary human umbilical vein endothelial cells by liposome-mediated gene transfection, Culture of human umbilical vein endothelial cells cultured for 25 passages. Immunocytochemical staining showed positive for IECVIII. Flow cytometry detection of type I bean curd agglutinin binding rate of 97%. Chromosomal analysis shows that the structure and number of chromosomes in IEC cells have changed. In vitro experiments show that, IEC has the ability to support hematopoiesis
