目的探讨肿瘤细胞膜组成及流动性在癌转移中的作用及机制。方法以具有不同转移表型的人肺腺癌细胞系Anip及AGZY为对象,采用32P标记法及薄膜层层析和气相层析技术分析两个细胞系的膜脂及脂肪酸组成,并用荧光偏振技术测定两者的膜脂流动性。结果两者在磷脂和脂肪酸及膜流动性上均有显著不同,转移性较高的Anip细胞质膜含有较多的磷脂酸(PA)和磷脂酰肌醇(PI)及较少的肉豆蔻酸(myristicacid),并具有较低的流动性。结论癌细胞的转移能力与膜脂肪酸组成有密切关系。
Objective To explore the role and mechanism of tumor cell membrane composition and fluidity in cancer metastasis. Methods Human lung adenocarcinoma cell lines Anip and AGZY with different metastatic phenotypes were studied. The lipid and fatty acid composition of the two cell lines were analyzed by 32P labeling and thin layer chromatography and gas chromatography. Both membrane lipid fluidities were measured. The results showed that there were significant differences in phospholipids, fatty acids, and membrane fluidity. The highly metastatic Anip cell plasma membrane contained more phosphatidic acid (PA) and phosphatidylinositol (PI) and less myristic acid ( Myristicacid), and has low fluidity. Conclusion The metastatic capacity of cancer cells is closely related to the fatty acid composition of the membrane.
