目的以碱性磷酸酶(AKP)为指标,采用正交模拟法分析护骨胶囊十味药不同模拟组合的药效。方法将护骨胶囊十味药按照正交表,分为12个组。血清药理学实验获取12个组的含药血清,AKP法检测各组血清对成骨细胞分化的影响。通过正交模拟法与非线性混合效应模型,分析各组分在复方背景下的重要程度和相互作用。结果护骨胶囊各组分在复方中的药效贡献值顺序为淫羊藿>山药>巴戟天=熟地黄>杜仲>骨碎补>制首乌>龟甲>当归>续断。最佳药效组合为淫羊藿、巴戟天、骨碎补、杜仲、制首乌、熟地黄、山药。结论采用正交模拟法分析的结果提示护骨胶囊十味药中淫羊藿对成骨细胞分化起主要的作用,续断、当归及龟甲对成骨细胞分化有抑制作用。
OBJECTIVE To determine the pharmacodynamic effects of different combinations of Shiwei capsule and Shiwei drug by using orthogonal test to determine the activity of alkaline phosphatase (AKP). Methods According to the orthogonal table, the capsule of Shiwei capsule was divided into 12 groups. Serum pharmacology experiments obtained serum containing 12 groups, AKP method to detect the effect of each group of serum on osteoblast differentiation. Through the orthogonal simulation and non-linear mixed effects model, the importance and interaction of each component in the compound background were analyzed. Results The order of contribution of each component of Prescription for Caregive Capsules to compound was Epimedium> Chinese yam> Morinda officinalis = Rehmanniae radix. The best combination of medicinal herbs Epimedium, Morinda, Drynaria, Eucommia, Radix, Rehmannia, Chinese yam. Conclusion The results of orthogonal simulation suggest that Epimedium sinense in the capsule of ten drugs plays a major role in the differentiation of osteoblasts, and the decoction of Angelica and Angelica sinensis can inhibit the differentiation of osteoblasts.
