目的 观察已受雌、孕激素影响的人子宫内膜细胞在一氧化氮(NO)作用下的细胞生理变化。方法 取人分泌中期子宫内膜,体外原代培养24~48 h,用钙荧光探剂Fluo-3-AM 负荷钙离子,采用激光扫描共聚焦显微镜测定加入一氧化氮合酶抑制剂(L-NAME)后,子宫内膜细胞内游离钙离子浓度([Ca2+]i)的动态变化。结果 加入L-NAME后,腺上皮细胞和间质细胞[Ca2+]i立即开始升高,400 s后上升至高峰,900 s时稍有下降,然后继续升高并保持在高水平。同时发现腺上皮细胞和基质细胞对L-NAME的反应不同。结论 在雌、孕激素作用的基础上,NO通过改变子宫内膜细胞[Ca2+]i进行信号转导,实现其生理效应。
Objective To observe the changes of cell physiology in endometrial cells which have been affected by estrogen and progesterone under the action of nitric oxide (NO). Methods The human endometrium was secreted in vitro. Primary cultured in vitro for 24 ~ 48 h, Calcium ion was loaded on Fluo-3-AM using calcium fluorescence probe. Laser confocal microscopy was used to observe the effects of nitric oxide synthase inhibitor (L- NAME), the endometrial intracellular free calcium concentration ([Ca2 +] i) dynamic changes. Results After adding L-NAME, [Ca2 +] i of glandular epithelial cells and interstitial cells began to rise immediately, peaked at 400 s, decreased slightly at 900 s, then continued to increase and remained at a high level. Also found that glandular epithelial cells and stromal cells respond differently to L-NAME. Conclusion Based on the effects of estrogen and progesterone, NO modulates the signal transduction of [Ca2 +] i in endometrial cells and realizes its physiological effects.
