研究β-酪啡肽-7(β-casomorphin-7,β-CM-7)对链脲佐菌素诱导的糖尿病大鼠肾损伤的保护作用.结果显示:(1)β-酪啡肽-7降低糖尿病大鼠的采食量、饮水量和空腹血糖值,提高糖尿病大鼠的体重,但是差异不显著(P>0.05).(2)β-酪啡肽-7显著降低糖尿病大鼠尿糖、尿蛋白、血清肌酐、血清尿素氮、肾脏指数和血清中转化生长因子-β1(TGF-β1)的含量;Masson染色结果显示,β-酪啡肽-7治疗显著降低糖尿病大鼠肾脏的纤维化程度.(3)RT-RCR结果显示,β-酪啡肽-7显著降低糖尿病大鼠肾脏TGF-β1、Ⅰ型和Ⅲ型胶原蛋白基因的表达.Western印迹显示,β-酪啡肽-7显著降低糖尿病大鼠肾脏Ⅰ型和Ⅲ型胶原蛋白的表达.上述结果表明,β-酪啡肽-7能够减缓糖尿病大鼠肾脏功能和肾脏纤维化程度,其机制可能与降低TGF-β1含量、减少胶原蛋白在肾脏的沉积有关.
To investigate the protective effect of β-casomorphin-7 (β-CM-7) on renal injury induced by streptozotocin in streptozotocin-induced diabetic rats.Results: (1) 7 reduced the intake, water intake and fasting blood glucose of diabetic rats, and increased the body weight of diabetic rats, but the difference was not significant (P> 0.05). (2) β -Morphine-7 significantly reduced the urine of diabetic rats Sugar, urinary protein, serum creatinine, serum urea nitrogen, kidney index and serum TGF-β1 content; Masson staining results showed that β-caspase-7 treatment significantly reduced the diabetic rat kidney (3) The results of RT-RCR showed that β-caspase-7 significantly decreased the expression of TGF-β1, type Ⅰ and type Ⅲ collagens in diabetic rats.Western blotting showed that β-cascanin -7 significantly reduced the expression of type I and type III collagen in kidney of diabetic rats.The above results show that beta-caspase-7 can reduce the renal function and the degree of renal fibrosis in diabetic rats, which may be related to the decrease of TGF- Content, reduce the deposition of collagen in the kidney.
