目的 比较人粘液表皮样癌耐药细胞株 (MEC -1/ 5 -FU )及其亲本细胞 (MEC -1)的线粒体数量和分布特点。方法 利用免疫荧光细胞化学染色结合激光扫描共聚焦显微镜、透射电镜观察两种细胞的线粒体分布及表达特征 ;细胞计数法观察细胞生长曲线及群体倍增时间。结果 MEC -1和MEC -1/ 5 -FU细胞增殖活跃 ,细胞群体倍增时间分别为 3 0h和 2 8.8h ;MEC -1/ 5 -FU细胞线粒体的数量与分布较MEC -1细胞数量更多 ,多集中分布于细胞质内 ,且集中分布在粗面内质网附近。MEC -1/ 5 -FU细胞线粒体外形较不规则 ,嵴较发达。结论 MEC -1和MEC -1/ 5 -FU细胞线粒体数量和分布的不同以及线粒体嵴的数量、形态及排列方式、线粒体外形、嵴的发达程度等的差异均提示二者可能在能量消耗、细胞代谢及蛋白质合成功能方面存在差异
Objective To compare the number and distribution of mitochondria in human mucoepidermoid carcinoma cell line MEC-1/5-FU and its parental cells (MEC-1). Methods Immunofluorescence cytochemistry combined with laser scanning confocal microscopy and transmission electron microscopy were used to observe the mitochondria distribution and expression characteristics of the two cells. Cell growth curve and population doubling time were observed by cell counting. RESULTS: The proliferation of MEC-1 and MEC-1/5-FU cells was active with a population doubling time of 30 h and 28.8 h, respectively. The number and distribution of mitochondria in MEC-1/5 -FU cells were more than those in MEC- , Mostly concentrated in the cytoplasm, and concentrated in the vicinity of the rough endoplasmic reticulum. MEC-1/5 -FU mitochondrial more irregular shape, ridges more developed. Conclusions The differences in mitochondrial numbers and distributions, number, morphology and arrangement of mitochondrial cristae, mitochondrial shape and cristae development in MEC-1 and MEC-1/5-FU cells suggest that the two may play roles in energy expenditure, There are differences in metabolism and protein synthesis
