【目的】研究不同凝胶基质对凝胶剂中左旋肉碱体外经皮渗透的影响,并评价凝胶剂作为左旋肉碱透皮给药载体的可行性。【方法】制备不同基质的左旋肉碱凝胶,采用改良的Franz扩散池,用离体大鼠皮肤进行体外经皮渗透实验,以HPLC法检测接受液中左旋肉碱含量并计算累积渗透量及稳态透皮速率。【结果】以2%羧甲基纤维素钠为基质的左旋肉碱累积透过量最大(7.635 mg/cm~2),透皮速率常数(J)为0.2308 mg·(cm~2·h)~(-1),均优于甲基纤维素和卡波姆。【结论】选用2%羧甲基纤维素钠作为左旋肉碱凝胶剂的基质较好。
【Objective】 The purpose of this study was to investigate the effects of different gel matrix on the transdermal penetration of L-carnitine in the gel and to evaluate the feasibility of the gel as a transdermal delivery vector for L-carnitine. 【Method】 L-carnitine gels with different matrices were prepared. The modified Franz diffusion cells were used to in vitro transdermal permeation test of rat skin. The content of L-carnitine in the solution was measured by HPLC and the cumulative permeation and Steady state transdermal rate. 【Result】 The results showed that the transdermal rate constant (J) was 0.2308 mg · cm ~ (2 · h) ~ (-2) at the highest cumulative concentration of 2-carboxymethylcellulose sodium as substrate (-1), both better than methylcellulose and carbomer. 【Conclusion】 The selection of 2% sodium carboxymethyl cellulose as L-carnitine gel matrix is better.
