目的探讨参佛胃康汤剂对胃癌细胞的抗瘤作用及其机制。方法予昆明种小鼠皮下接种小鼠胃癌细胞MFC造模,将造模后的小鼠随机分为模型组和参佛胃康高、中、低剂量组;在肿瘤移植后的第1天ig给予实验组小鼠参佛胃康汤剂,在肿瘤移植后第61天处死各组小鼠。定期检测小鼠的生存状态、肿瘤大小及重量;采用TUNEL法和流式细胞仪测定肿瘤细胞的凋亡指数;采用免疫组化法检测肿瘤组织中血管内皮生长因子(VEGF)、STAT3、缺氧诱导因子-1α(HIF-1a)的表达。结果与模型组比较,参佛胃康组小鼠肿瘤的大小和重量明显受到抑制,其凋亡指数显著增加;肿瘤组织中VEGF、STAT3、HIF-1a的表达下调。结论参佛胃康汤剂可能通过诱导肿瘤细胞凋亡和下调VEGF、STAT3、HIF-1a等信号转导蛋白的表达来发挥其抗肿瘤作用。
Objective To investigate the anti-tumor effect of Shenfu Weikang decoction on gastric cancer cells and its mechanism. Methods Kunming mice were inoculated subcutaneously in mice with gastric cancer MFC model, the mice were randomly divided into model group and Shenweikang high, medium and low dose group; on the first day after tumor transplantation ig The mice in the experimental group were given Fuweikang decoction, and the mice in each group were sacrificed on the 61st day after tumor transplantation. The survival status, tumor size and weight of mice were detected regularly. The apoptosis index of tumor cells was determined by TUNEL and flow cytometry. The expressions of VEGF, STAT3, Inducible factor-1α (HIF-1a) expression. Results Compared with the model group, the size and weight of the tumor of Shenfo Weike group were significantly inhibited, and the apoptosis index of the model group was significantly increased. The expression of VEGF, STAT3 and HIF-1a was down-regulated in the tumor tissue. Conclusion Shenfu decoction may exert its anti-tumor effect by inducing apoptosis of tumor cells and down-regulating the expression of signal transducers such as VEGF, STAT3 and HIF-1a.
