目的:研究一种环保、高效的苦参生物碱分离技术。方法:以苦参总碱吸附量和解吸率为指标,从大范围筛选树脂,并以选出的树脂进一步优化其吸附与解吸的条件。结果:所选出的树脂为X-5型非极性大孔树脂;优化的吸附条件为上柱液pH10、盐离子浓度1mol·L-1,吸附流速6BV·h-1,总生物碱的吸附量可达9.74mg·mL-1;优化的洗脱条件为60%乙醇(pH1)洗脱,解吸率可达95.5%。结论:X-5型树脂能直接从苦参渗漉液中快速吸附分离生物碱,吸附量大、解吸容易,并对环境、产品无污染,极具开发成绿色工艺潜力。
Objective: To study a kind of environment-friendly and efficient separation technique of alkaloids from matrine. Methods: Based on the total alkali absorption and desorption rate of Sophora flavescens, the resin was screened from a wide range and the conditions of adsorption and desorption were further optimized with the selected resin. Results: The selected resin was X-5 nonpolar macroporous resin. The optimal adsorption conditions were as follows: the upper column liquid pH10, the salt ion concentration 1mol·L-1, the adsorption flow rate 6BV · h-1, the total alkaloid The adsorption capacity was up to 9.74 mg · mL-1. The optimal elution conditions were 60% ethanol (pH1) and the desorption rate reached 95.5%. Conclusion: X-5 resin can directly adsorb and separate alkaloids directly from Sophora flavescens percolate. It has the advantages of large adsorption capacity, easy desorption, no pollution to the environment and products, and great potential for developing green technology.
