目的 研究心房颤动 (AF)患者内向整流钾电流 (Ik1 )重构的分子基础。方法 将 4 6例风湿性心瓣膜病接受换瓣手术患者分为三组 ,窦性心律组 (SR组 ) 18例 ,阵发性房颤组 (PAF) 7例 ,慢性房颤组 (CAF) 2 1例。应用半定量逆转录 -聚合酶链反应 (RT- PCR)技术 ,检测心房组织 Kir2 .1m RNA表达。结果 和窦性心律组相比 ,Kir2 .1m RNA在慢性房颤组中的表达明显增加 (P<0 .0 1)。结论 风湿性心瓣膜病伴房颤患者 Kir2 .1m RNA表达水平的增加可能是 IK1 上调的分子基础。
Objective To study the molecular basis of remodeling of inward rectifier potassium current (Ik1) in patients with atrial fibrillation (AF). Methods Forty - six patients with rheumatic valvular heart disease undergoing valve replacement surgery were divided into three groups: 18 patients in sinus rhythm group (SR group), 7 patients in paroxysmal atrial fibrillation group (PAF), 7 patients in chronic atrial fibrillation group (CAF) 2 1 case. Semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression of Kir2.1 mRNA in atrial tissue. Results Compared with sinus rhythm group, the expression of Kir2.1 mRNA in chronic atrial fibrillation group was significantly increased (P <0.01). Conclusion The increased expression of Kir2.1mRNA in patients with rheumatic valvular heart disease and atrial fibrillation may be the molecular basis of IK1 up-regulation.
