用刀豆球蛋白A(concanavalin A,Con A)预先处理的异种移植神经,对神经再生的影响。含Con A50mg的Ringer液1 000ml(4℃)灌注日本大耳兔2h后,取出兔的胫神经。麻醉Wistar大白鼠,切除其右坐骨神经5mm,移植入8mm长,经Con A处理的兔胫神经。术后4、8、12周,可见再生神经纤维通过近端吻合部,长入异种移植神经,继而越过远端吻合部,向远段坐骨神经生长。移植神经段和远段坐骨神经中再生神经纤维成束或散在分布,再生的无髓纤维散在于有髓纤维之间。8周以后,大鼠手术侧的腓肠肌乙酰胆碱酯酶(AChE)呈阳性,再生神经终末纤维与运动终板相连。手术侧爪部皮肤的真皮和表皮,出现再生神经纤维和游离感觉神经末梢。Con A预先处理兔神经,有利于宿主的再生神经纤维能通过异种移植神经向远段生长,重新支配靶器官。
Effects of preconditioned xenograft nerves with concanavalin A (Con A) on neural regeneration. Rabbit tibial nerves were removed after 1 000 ml (4 ° C) Ringer’s solution containing Con A 50 μg was perfused into Japanese white rabbits for 2 h. Wistar rats were anesthetized, their right sciatic nerve was excised 5mm, transplanted into 8mm long, con A-treated rabbit tibial nerve. At 4, 8 and 12 weeks after operation, regenerating nerve fibers were seen to grow into the distal sciatic nerve through the proximal anastomosis and into the xenograft nerve, then over the distal anastomosis. The regenerated nerve fibers in the graft and distal sciatic nerves are bundled or scattered, and the regenerated non-myelinated fibers are scattered among the myelinated fibers. After 8 weeks, AChE was positive in the operation side of the rat and the regenerated nerve terminal fibers were connected with the motor endplate. The dermis and epidermis of the skin on the side of the operation side claw appear regenerative nerve fibers and free sensory nerve endings. Con A pretreatment of the rabbit nerve is conducive to the host regenerative nerve fibers can grow through the distal part of the xenograft to re-target the target organ.
