目的 :研究 1,2 5 -二羟维生素D3 [1α,2 5 -dihydroxyvitaminD3 ,1α,2 5 (OH) 2 D3 ]对人成骨细胞膜型基质金属蛋白酶 - 1(membrane -typematrixmetalloproteinase - 1,MT1-MMP)表达的影响 ,探讨 1α,2 5 (OH) 2 D3 调节骨代谢作用机制。方法 :人成骨细胞用 1α ,2 5 (OH) 2 D3 干预。Western杂交检测MT1-MMP蛋白质表达。MMP - 2活性用ELISA检测。结果 :观察到 1α ,2 5 (OH) 2 D3 诱导成骨细胞MT1-MMP表达呈剂量依赖性 (均P <0 0 5 )。 1α,2 5 (OH) 2 D3 干预 12~ 2 4h促进MT1-MMP表达 (均P <0 0 5 )。 1α,2 5 (OH) 2 D3 促进MMP - 2激活呈剂量依赖性 (均P <0 0 5 )。结论 :由于MT1-MMP在骨重建过程中起着维持骨形成的关键作用 ,1α ,2 5(OH) 2 D3 可通过诱导成骨细胞MT1-MMP表达促进骨形成。此可能为 1α,2 5 (OH) 2 D3 治疗骨质疏松症机理之一。
AIM: To investigate the effect of 1,25 - dihydroxyvitamin D3 [1α, 2 5 (OH) 2 D3] on the expression of membrane type matrix metalloproteinase - 1 (MT1 - MMP) expression, explore the mechanism of 1α, 25 (OH) 2 D3 in regulating bone metabolism. Methods: Human osteoblasts were treated with 1α, 25 (OH) 2 D3. Western blot was used to detect MT1-MMP protein expression. MMP-2 activity was detected by ELISA. Results: MT1-MMP expression induced by 1α, 2 5 (OH) 2 D3 in osteoblasts was dose-dependent (all P <0.05). 1α, 2 5 (OH) 2 D3 could promote the expression of MT1-MMP in 12 ~ 24 hours (all P <0 05). 1α, 2 5 (OH) 2 D3 promoted the activation of MMP - 2 in a dose - dependent manner (all P <0 05). Conclusion: 1α, 25 (OH) 2 D3 can promote bone formation by inducing MT1-MMP expression in osteoblasts because MT1-MMP plays a key role in bone remodeling. This may be 1α, 25 (OH) 2 D3 treatment of osteoporosis one of the mechanisms.
