目的 研究从转录水平确定晶状体上皮细胞是否能够合成纤溶酶原激活物抑制物 1(PAI 1)蛋白酶。方法 体外培养的牛晶状体上皮细胞 ,当细胞生长融合后 ,抽提RNA ;采用Nothernblot分析技术 ,对抽提得到的牛晶状体上皮细胞RNA用特异的cDNA探针进行检测。结果 在牛晶状体上皮细胞的基因转录产物中存在有PAI 1mRNA的表达 ,所表达的PAI 1mRNA碱基长度为 2 .3kb。结论 牛晶状体上皮细胞能够分泌PAI 1蛋白酶 ;该蛋白酶可能对白内障囊外摘出术后房水内纤维蛋白反应的发生起到重要的作用
OBJECTIVE: To determine whether lens epithelial cells can synthesize plasminogen activator inhibitor 1 (PAI 1) protease at the transcriptional level. Methods Bovine lens epithelial cells cultured in vitro were extracted from RNA after the cells grew and fused. Nothernblot assay was used to detect the RNA of extracted bovine lens epithelial cells with specific cDNA probes. Results The expression of PAI 1 mRNA existed in the gene transcription products of bovine lens epithelial cells, and the base length of PAI 1 mRNA expressed was 2.3 kb. Conclusions Bovine lens epithelial cells are capable of secreting PAI 1 protease; this protease may play an important role in the fibrin response in aqueous humor after extracapsular cataract extraction
