目的研究聚乙二醇-6000(polyethylene glycol-6000,PEG-6000)胁迫对丹参毛状根丹参酮类成分积累的影响。方法用发根农杆菌ATCC15834诱导生产丹参毛状根,经20 d悬浮振荡培养后,分别采用1.2%、2.0%、5.5%、10%的PEG-6000处理,并用HPLC法测定PEG-6000处理1周后丹参毛状根中丹参酮I、隐丹参酮、二氢丹参酮I、丹参酮IIA的量。结果PEG-6000能够抑制丹参毛状根的生长,PEG-6000为1.2%、2.0%、5.5%和10%处理条件下,丹参毛状根干质量分别减少为对照组的75.1%、83.0%、76.2%、76.1%;PEG-6000处理能够显著提高丹参毛状根中4种丹参酮的量,2.0%、5.5%的PEG-6000能够显著促进丹参酮I量的提高,1.2%PEG-6000能够显著提高隐丹参酮量的;二氢丹参酮I的量以2.0%PEG-6000处理条件下最高;丹参酮IIA的量以5.5%PEG-6000处理条件下最高。结论 PEG-6000能显著促进丹参毛状根中丹参酮类成分的积累。
Objective To investigate the effects of polyethylene glycol-6000 (PEG-6000) stress on the accumulation of tanshinones in hairy root of Salvia miltiorrhiza. Methods Hairy roots of Salvia miltiorrhiza were induced by Agrobacterium rhizogenes ATCC15834 and cultured in suspension for 20 days. The cells were treated with PEG-6000 at 1.2%, 2.0%, 5.5% and 10%, respectively. PEG-6000 treatment 1 Weeks after Salvia hairy root tanshinone I, cryptotanshinone, dihydrotanshinone I, tanshinone IIA amount. Results PEG-6000 could inhibit the growth of hairy roots of Salvia miltiorrhiza. The dry weight of hairy roots of Salvia miltiorrhiza decreased to 75.1% and 83.0% of that of the control group under PEG-6000 treatment of 1.2%, 2.0%, 5.5% and 10% 76.2% and 76.1%, respectively. PEG-6000 treatment significantly increased the amount of 4 tanshinones in the hairy roots of Salvia miltiorrhiza. 2.0% and 5.5% PEG-6000 significantly increased the amount of tanshinone I and 1.2% PEG-6000 significantly increased The amount of cryptotanshinone was the highest; the amount of dihydrotanshinone I was the highest under 2.0% PEG-6000 treatment; the amount of tanshinone IIA was the highest under 5.5% PEG-6000 treatment. Conclusion PEG-6000 can significantly promote the accumulation of tanshinones in hairy roots of Salvia miltiorrhiza.
