目的:探讨抗肿瘤单体没食子酸(GA)对正常人肝细胞(LO2)的体外细胞毒性。方法:采用MTT实验、流式细胞术、Hoechst-33258荧光染色法和电镜检测。结果:GA作用24h后,0,6.25,12.5,25,50,100,200μmol/L用药组出现不同程度的细胞增殖抑制现象,抑制率分别为0,18.2%,25.5%,42.0%,42.5%,51.8%,60.6%;流式细胞仪检测0,25,50,100μmol/L用药组凋亡率分别为1.37%,4.63%,4.64%,12.53%;荧光染色和电镜检测提示100μmol/L组细胞有形态改变,细胞核固缩、碎裂。结论:抗肿瘤单体GA对LO2细胞增殖也有抑制作用,并能诱导LO2细胞凋亡。
Objective: To investigate the in vitro cytotoxicity of anti-tumor monomeric gallic acid (GA) on normal human hepatocytes (LO2). Methods: MTT assay, flow cytometry, Hoechst-33258 fluorescence staining and electron microscopy. Results: After treated with GA for 24 h, the proliferation inhibition rates of 0, 6.25, 12.5, 25, 50, 100 and 200 μmol / L groups were all different. The inhibitory rates were 0,18.2%, 25.5%, 42.0%, 42.5% and 51.8% , 60.6% respectively. The apoptotic rates of the treated groups were 1.37%, 4.63%, 4.64% and 12.53% respectively by flow cytometry. Fluorescence staining and electron microscopy suggested that the morphological changes of cells in 100μmol / L group , Nuclear condensation, fragmentation. Conclusion: Anti-tumor monomeric GA can also inhibit the proliferation of LO2 cells and induce the apoptosis of LO2 cells.
