目的探讨松仁油对脂代谢相关基因表达的影响。方法分别以基础饲料、高脂饲料(20%猪油和80%基础饲料HF)及高脂饲料加松仁油(20%松仁油和80%高脂饲料SHF)饲喂小鼠,记录体重调整饲喂量,于45d和90d时取睾周脂肪,以RT-PCR法测定酰基CoA合成酶(ACS)、脂肪酸转运蛋白(FATP)、激素敏感脂肪酶(HSL)、过氧化物酶体增殖剂激活受体-γ(PPAR-γ)mRNA的表达水平。结果45d时,SHF组与基础组比,ACSmRNA、FATPmRNA、HSLmRNA和PPAR-γmRNA的表达水平分别增加了24.2%、8.8%、10.9%、21.6%。90d时,SHF组与基础组比,ACSmRNA、FATPmRNA、HSLmRNA和PPAR-γmRNA的表达水平分别增加了19.6%、25.7%、60.1%、13.1%;HF组与基础组比ACSmRNA、FATPmRNA、HSLmRNA和PPAR-γmRNA的表达水平分别增加了27.9%、41.7%、51.1%、23.9%。结论松仁油在调节脂质代谢方面具有积极的意义。
Objective To investigate the effect of pine nut oil on lipid metabolism related gene expression. Methods Mice were fed with basal diet, high fat diet (20% lard and 80% basal diet HF) and high fat diet with pine nut oil (20% pine nut oil and 80% SHF) Peripheral blood was collected at 45 days and 90 days. Acyl CoA synthetase (ACS), fatty acid transporter (FATP), hormone sensitive lipase (HSL) and peroxisome proliferator-activated Receptor-γ (PPAR-γ) mRNA expression levels. Results At 45 days, the expression of ACS mRNA, FATP mRNA, HSL mRNA and PPAR-γ mRNA increased by 24.2%, 8.8%, 10.9% and 21.6% in SHF group and basal group, respectively. At 90 days, the expression of ACS mRNA, FATP mRNA, HSL mRNA and PPAR-γ mRNA increased by 19.6%, 25.7%, 60.1% and 13.1% in SHF group and basal group, respectively. γ mRNA expression levels increased by 27.9%, 41.7%, 51.1%, 23.9%. Conclusion Pine nut oil in the regulation of lipid metabolism has a positive meaning.
