环硫雄醇(Epitiostanol)学名:2α,3α-环硫-5α-雄甾-17β醇,据报道有抗癌作用。最近我们在国内首次人工合成环硫雄醇纯品,并以免疫抑制小鼠及人乳腺癌细胞株(BCap-37)和子宫颈癌细胞株(HeLa)为模型,进行了抗癌活性的研究。实验方法与结果一、剂量反应试验:BCap-37及HeLa细胞传代并各接种21瓶(3×10~5细胞/瓶),随机分成7组,每3瓶一组;置37℃温箱培养24小时后更换新鲜培养液,分别加入0、50、100、250、500、1000μg/ml的环硫雄醇及50ml蒸馏水;在37℃温箱内作用12小时后以苔盼蓝排除法计算各剂量组的细胞存活率,取存活率50%对应剂量作为实验剂量。细胞存活率=(存活细胞数+修正数)/(3×10~5)×100% (修正数=3×10~5-空白组细胞存活数)本实验结果取500μg/ml为实验剂量。
Epitiostanol scientific name: 2α, 3α-episulfan-5α-androstan-17β-ol, reported to have anti-cancer effects. In the dose group, the cell survival rate was 50% of the survival rate as the experimental dose. Cell survival rate = (survival cell number + correction number)/(3 x 10~5) x 100% (correction number = 3 x 10 ~ 5 - number of cells in the blank group) The experimental result was 500 μg/ml.
