目的:探讨肿瘤坏死因子相关凋亡诱导配体(TRAIL)对人乳腺癌(MCF-7)、宫颈癌(Hela)、卵巢癌(SKOV-3)细胞体外抗肿瘤的杀伤作用。方法:通过MTT法、流式细胞术检测重组人TRAIL蛋白对3种肿瘤细胞和人胚肾(HEK-293)正常细胞系的作用和诱导凋亡情况。结果:一定浓度的TRAIL可有效抑制MCF-7、SKOV-3、Hela细胞,其生长抑制呈剂量依赖。TRAIL对SKOV-3细胞最敏感,用2.5 mg/L TRAIL作用SKOV-3,12h后发生凋亡,其细胞凋亡率呈时间依赖。结论:重组人TRAIL对人乳腺癌、宫颈癌、卵巢癌细胞系具有生长抑制作用,并能够诱导SKOV-3细胞凋亡。
Objective: To investigate the anti-tumor effect of tumor necrosis factor related apoptosis inducing ligand (TRAIL) on human breast cancer (MCF-7), cervical cancer (Hela) and ovarian cancer (SKOV-3) Methods: MTT assay and flow cytometry were used to detect the effect of recombinant human TRAIL protein on normal tumor cell line and normal human embryonic kidney (HEK-293) cell line and the induction of apoptosis. Results: TRAIL could inhibit the proliferation of MCF-7, SKOV-3 and Hela cells in a dose-dependent manner. TRAIL was the most sensitive to SKOV-3 cells. After being treated with 2.5 mg / L TRAIL for 2 days, SKOV-3 cells were apoptosis induced by TRAIL. The apoptosis rate of SKOV-3 cells was time-dependent. Conclusion: Recombinant human TRAIL can inhibit the growth of human breast cancer, cervical cancer and ovarian cancer cell lines and induce the apoptosis of SKOV-3 cells.
