以西瓜抗ZYMV-CH(小西葫芦黄花叶病毒中国株系,Zucchini yellow mosaic virus-CHINA)野生种质P.I.595203和感病自交系98R为试材构建F2群体和F2:3群体,采用BSA法筛选与抗病基因zym-CH连锁的分子标记,并利用以F2S8群体(P.I.296341-FR×97103)构建的西瓜参考遗传图谱进行zym-CH的比较定位。试验结果表明:在参考遗传图谱已经定位的RAPD和SSR标记中,RAPD标记b13-1400与zym-CH的遗传距离为25.6cM;SSR标记MCPI-14和MCPI-26与zym-CH的遗传距离分别为57cM和87cM,比较分析发现这些标记在两个图谱上的分布呈共线性关系。根据zym-CH与标记b13-1400、MCPI-14和MCPI-26之间的遗传关系,将西瓜抗病毒病基因zym-CH初步定位在西瓜参考遗传图谱3号连锁群105cM处。
The F2 population and the F2: 3 population were constructed from the wild germplasm PI595203 and susceptible inbred line 98R of ZYMV-CH (Zucchini yellow mosaic virus-CHINA) The molecular markers linked to the resistance gene zym-CH were screened and the zym-CH loci were mapped using the reference genetic map of watermelon constructed with the F2S8 population (PI296341-FR × 97103). The results showed that the genetic distance between RAPD marker b13-1400 and zym-CH was 25.6cM in the RAPD and SSR markers which were mapped by reference genetic map. The genetic distance between MCP-14 and MCP-26 and zym-CH was 57cM and 87cM, comparative analysis found that the distribution of these markers in the two maps were co-linear relationship. According to the genetic relationship between zym-CH and markers b13-1400, MCPI-14 and MCPI-26, the zym-CH gene of watermelon antiviral was initially located at 105 cM of the linkage map of the reference genetic map of watermelon.
