用塑料袋制菌种,常用棉塞或扎线封袋口,前者在灭菌时棉塞易受潮而增加污染率,后者则由于通气差而导致后期菌丝长速慢、纤细长势不良。我们采用厦门市厦青塑料制品厂生产的无棉盖体塑料双套环制菌种,取得了良好的效果。 1 无棉盖体结构 分套环和盖两部分。盖中央有一直径1.1cm的孔,下垫一层无棉盖体,盖体下衬一张有6个小孔塑料薄片。使用时在套环上盖上盖子,压紧即可。 2 试验方法 基质配方为木屑78.5%,麸皮20%,石灰0.5%。共试验两批。第一批封口材料:a.无棉套盖;b.套环加棉塞。15×30cm的聚丙烯袋,高压灭菌。第二批封口材料:c.无棉套盖,d.棉线扎紧袋口。17×34cm的低压聚乙烯袋,常压灭菌。分别于1998年6月12、16日接毛木耳193,每组60袋,27~31℃培养。菌丝封面后隔7天测菌丝长速,各测2次,记录污染袋数。 3 试验结果 ①菌丝长速:接种后7~8天,菌丝封面,各组差异不显著。采用无棉盖体套盖的菌丝日均长速与用棉塞封口的差异不显著,但显著快于用棉线扎袋口
