本文阐述体外由植物血凝素(PHA)激发的人外周血淋巴细胞化学发光的实验条件,并研究叠氮钠、松胞素B、秋水仙胺对淋巴细胞化学发光峰值的影响。结果表明,叠氮钠(10,20,40mmol/L。)能够通过抑制淋巴细胞膜NADPH氧化酶氧化磷酸化而减弱淋巴细胞化学发光(P<0.01)。松胞素B(1,5,10μg/ml)和秋水仙胺(1,2,4μg/ml)分别破坏淋巴细胞的微管、微丝而抑制淋巴细胞化学发光(P<0.01)。由此可见,NADPH氧化酶氧化磷酸化,细胞骨架在淋巴细胞化学发光中起着重要的作用。
In this paper, the experimental conditions of chemiluminescence of human peripheral blood lymphocytes stimulated by phytohemagglutinin (PHA) in vitro and the effects of sodium azide, cytochalasin B and colchicine on the chemiluminescence peak of lymphocytes were studied. The results showed that sodium azide (10,20,40 mmol / L) attenuated lymphocyte chemiluminescence (P <0.01) by inhibiting the oxidative phosphorylation of NADPH oxidase on lymphocyte membrane. Cytokines (1, 5, 10μg / ml) and colchicine (1,2,4μg / ml) respectively destroys the microtubules and microfilaments of lymphocytes and inhibits lymphocyte chemiluminescence (P <0.01). Thus, NADPH oxidase oxidative phosphorylation, cytoskeleton in lymphocyte chemiluminescence plays an important role.
