目的:探讨补肾明目冲剂延缓和预防机体组织细胞生理衰老的机理。方法:选用自然衰老的16月龄Wistar大鼠,用补肾明目冲剂灌胃30天,维生素E为阳性对照组,灌胃结束后,取大鼠心、脑、晶状体蛋白和视网膜脉络膜组织。用丙二醛法测定脂质过氧化物(lipidperoxids,LPO)含量;用亚硝酸盐试剂盒检测超氧化物歧化酶(superoxidedismutase,SOD)活性;用DTNB法测定谷光甘肽过氧化物酶(glutathioneperoxidaseGSH-PX)活性;用钼酸铵显色法测定过氧化氢酶(catalaeCAT)活性;用Schif碱荧光法测定脂褐素含量。结果:补肾明目组大鼠晶状体蛋白、视网膜脉络膜组织LPO含量降低;SOD、GSH-PX和CAT酶活性增高;心肌和脑组织脂褐素含量降低。结论:补肾明目冲剂在抗氧化方面是有效的。
Objective: To explore the mechanism of Bushen Mingmu granules in delaying and preventing physiological aging of body cells. METHODS: Naturally aged 16-month-old Wistar rats were selected and fed with Bushen Mingmu granules for 30 days. Vitamin E was a positive control group. Rats’ heart, brain, lens protein, and retinal choroid tissue were collected after gastric filling. The content of lipid peroxides (LPO) was determined by the method of malondialdehyde; the activity of superoxide dismutase (SOD) was detected by nitrite kit; the glutathione peroxidase (GSH) was measured by DTNB method. -PX) activity; catalase activity was measured by ammonium molybdate coloration method; lipofuscin content was measured by Schif-base fluorescence method. RESULTS: The content of LPO in lens protein and retinal choroid tissue of the Kidney-tonming eyesight group decreased; the activities of SOD, GSH-PX and CAT enzymes increased; the content of lipofuscin in the heart and brain tissues decreased. Conclusion: Bushen Mingmu granules are effective in antioxidants.
