目的探讨海马微环境的变化对体外培养的放射状胶质细胞增殖和分化的影响。方法本实验在成功制备正常侧和切割穹隆海马伞侧海马提取液的基础上,分别使用正常侧和切割侧海马提取液,观察其在体外培养的放射状胶质细胞增殖和分化过程中所起的作用。结果在放射状胶质细胞增殖阶段,切割组与正常组及对照组相比,5-溴脱氧尿嘧啶核苷(BrdU)标记的增殖细胞比例明显提高(P<0.05);在放射状胶质细胞分化阶段,切割组与正常组及对照组相比,Tuj1阳性神经元数量明显增多(P<0.05)。结论切割穹隆海马伞侧海马提取液对大鼠放射状胶质细胞的增殖和向神经元的分化有明显促进作用,提示海马微环境对海马放射状胶质细胞的增殖和分化有重要影响。
Objective To investigate the effects of changes of hippocampus microenvironment on the proliferation and differentiation of radial glial cells cultured in vitro. Methods In this study, we successfully prepared hippocampal extracts from the normal and cut-fimbrial hippocampus using normal and excised hippocampal extracts respectively, and observed the proliferation and differentiation of hippocampal neurons cultured in vitro effect. Results In the stage of radial glial cell proliferation, the proportion of proliferating cells labeled with BrdU markedly increased (P <0.05) in the cutting group compared with the normal group and the control group. In the radial glial cell differentiation Compared with the normal group and the control group, the number of Tuj1 positive neurons in the cutting group increased significantly (P <0.05). Conclusions The hippocampus extract from hippocampus in the diencephalon can significantly promote the proliferation and differentiation of neurons in the astrocytes in hippocampus, suggesting that the hippocampal microenvironment has an important effect on the proliferation and differentiation of hippocampal radial glial cells.
