目的研究半夏及其炮制品清半夏、姜半夏、法半夏和京半夏总有机酸HPLC指纹图谱与止咳作用的相关性,建立半夏及其炮制品总有机酸与止咳作用的谱-效关系,以阐明止咳作用的物质基础。方法采用灰色关联度分析法,对半夏及其炮制品总有机酸HPLC指纹图谱的各共有峰峰面积与止咳的3个评价指标(咳嗽潜伏期、3 min内咳嗽次数及抑咳率)进行相关性分析,建立半夏及其炮制品总有机酸止咳作用的谱-效关系。结果半夏及其炮制品总有机酸对浓氨水致小鼠咳嗽有显著的抑制作用,与半夏止咳功效一致。在半夏及其炮制品总有机酸HPLC指纹图谱中,4、6(琥珀酸)、12号色谱峰所代表的化学成分与止咳作用具有较强的相关性,对止咳作用贡献较大,特别是4号色谱峰,峰面积大、关联度强。结论半夏及其炮制品总有机酸HPLC指纹图谱与其止咳作用具有相关性。通过谱-效关系研究,阐明了半夏及其炮制品止咳作用的物质基础,为其质量控制与评价提供了理论依据。
OBJECTIVE To study the correlation between HPLC fingerprints of total organic acids from Pinellia tuber, Pinellia ternata, Pinellia ternata and Pentaphylla chinensis, and its antitussive action, and establish the spectrum of total organic acids and antitussive action of Pinellia ternata and its processed products. Effect relationship to clarify the material basis of the role of cough. Methods The gray relational analysis method was used to correlate the peak areas of HPLC fingerprint of total organic acids of Pinellia ternata and its processed products with three evaluation indexes of cough (cough latency, cough frequency and suppression rate within 3 min) Sex analysis, the establishment of Pinellia and its processed products of organic acids cough synergistic effect spectrum. Results Pinellia and its processed products total organic acids on the cough caused by concentrated ammonia water has a significant inhibitory effect, consistent with the efficacy of Banxia cough. In the HPLC fingerprinting of total organic acids from Pinellia ternata and its processed products, the chemical constituents represented by 4,6 (succinic acid) and 12th peak have a strong correlation with the cough-relieving effect and have a great contribution to the cough-suppressing effect, especially Is No. 4 peak, peak area, the correlation is strong. Conclusion The HPLC fingerprints of total organic acids from Pinellia ternata and its processed products are correlated with its antitussive action. Through the study of spectrum-efficiency relationship, the material basis of the cough-inhibiting effect of Pinellia ternata and its processed products is clarified, which provides a theoretical basis for its quality control and evaluation.
