目的 观察钙离子在表皮的分布情况。 方法 裸鼠皮肤经丙酮处理后 ,采用离子捕获细胞化学法对裸鼠表皮钙离子的分布进行超微结构的定位。 结果 超微结构观察显示在正常的表皮中 ,基底层钙离子的浓度最低 ,由基底层到棘层钙离子的浓度逐渐增加 ,在颗粒层上部 ,钙离子的浓度最高。另外 ,裸鼠皮肤经丙酮处理后可导致表皮钙离子浓度梯度的丧失。 结论 表皮中存在一钙离子浓度梯度 ,离子细胞化学定位法是研究组织中钙离子分布的有效方法。
Objective To observe the distribution of calcium ions in the epidermis. Methods After the skin of nude mice were treated with acetone, the distribution of calcium ion in the epidermis of nude mice was localized by the ultrastructure using ion trapping cytochemistry. Results Ultrastructure observation showed that in the normal epidermis, the concentration of calcium ion in basal layer was the lowest, the concentration of calcium ion from basal layer to stratum spinosum increased gradually, and the calcium ion concentration was the highest in the upper part of granular layer. In addition, nude mice skin after acetone treatment can lead to the loss of epidermal calcium ion concentration gradient. Conclusion There is a gradient of calcium concentration in the epidermis, and ion cytochemical localization is an effective method to study the distribution of calcium in the tissue.
