本文用改良的焦锑酸盐法对兔肺和肾皮质钙离子(Ca~(2+))进行超微结构定位观察,在肺泡Ⅰ、Ⅱ型细胞、毛细血管内皮细胞和肾小管细胞胞质中均显示数量不等的Ca~(2+)沉淀产物,对这些Ca~(2+)沉淀产物用能谱仪进行X射线微区元素分析,表明含有钙的Kα峰和锑的Lα峰。用EGTA处理后,绝大多数Ca~(2+)的沉淀产物溶解。以上结果证实,用改良的焦锑酸盐法可在超微结构上原位显示Ca~(2+)。
In this study, the ultrastructure of calcium and calcium ions in the lung and renal cortex of rabbits was observed by modified pyroantimonate method. In the alveolar type Ⅰ and type Ⅱ cells, the capillary endothelial cells and the cytoplasm of renal tubular cells Showed different amounts of Ca2 + precipitates. X-ray microelements analysis of these Ca2 + precipitates by energy dispersive spectroscopy showed that the Kα peak containing calcium and the Lα peak of antimony were both observed. After treatment with EGTA, most of the precipitated product of Ca ~ (2+) was dissolved. The above results demonstrate that Ca ~ (2+) can be displayed in situ on the ultrastructure using a modified pyrophosphate method.
