目的:探讨生长因子受体结合蛋白2(Grb2)在胰腺癌中的表达及其意义。方法:分别采用real-time PCR和Western blot方法检测6种胰腺癌细胞株中Grb2基因和蛋白表达水平;采用组织芯片检测80例胰腺癌组织及癌旁胰腺组织中Grb2的表达情况,并分析Grb2表达状态与胰腺癌患者临床病理特征的关系。结果:Grb2的mRNA与蛋白在高侵袭力、低分化细胞株PANC-1、MIA-PaCa-2、AsPC-1中呈相对高表达,而在低侵袭力、高分化细胞株BxPC-3、SW1990、capan-2中呈相对低表达;Grb2在胰腺癌组织中均有表达,而在癌旁胰腺组织中不表达或低表达,统计分析显示,Grb2的高表达与肿瘤低分化以及淋巴结或者远处转移有关(均P<0.05)。结论:Grb2的表达与胰腺癌的分化程度和侵袭转移力密切相关,高表达者肿瘤恶性程度高,预后差。
Objective: To investigate the expression of growth factor receptor binding protein 2 (Grb2) in pancreatic cancer and its significance. Methods: The expression of Grb2 gene and protein in 6 pancreatic cancer cell lines were detected by real-time PCR and Western blot respectively. The expression of Grb2 in 80 pancreatic cancer tissues and adjacent normal pancreatic tissues was detected by tissue microarray. Grb2 Relationship between expression status and clinicopathological features in patients with pancreatic cancer. RESULTS: Grb2 mRNA and protein were relatively highly expressed in highly invasive and poorly differentiated cell lines PANC-1, MIA-PaCa-2 and AsPC-1. However, Grb2 mRNA and protein were highly expressed in low invasive and well differentiated cell lines BxPC-3 and SW1990 , While that of capan-2 was relatively low. Grb2 was expressed in pancreatic cancer tissues but not in adjacent pancreatic tissues. Statistical analysis showed that Grb2 overexpression was associated with poorly differentiated tumors and lymph nodes or distant sites Metastasis (all P <0.05). Conclusion: The expression of Grb2 is closely related to the degree of differentiation and invasion and metastasis of pancreatic cancer. The high expression of Grb2 is associated with high malignancy and poor prognosis.
