目的:考察党参总生物碱对神经生长因子诱导大鼠嗜铬神经瘤PC12细胞分化的影响。方法:用党参总生物碱在有或无神经生长因子(NGF)存在条件下培养PC12细胞,分析PC12细胞突起的延伸及相关蛋白激酶。结果:党参总生物碱自身不能诱导PC12细胞延伸突起,但在很低浓度NGF (2 μg/L)存在下,党参总生物碱能明显地诱导PC12细胞延伸出大量突起,增强有丝分裂原激活的蛋白激酶(MAPK)的磷酸化,而且这种增强作用能被有丝分裂原激活的蛋白激酶激酶特异抑制剂PD98059抑制。结论:党参总生物碱通过放大有丝分裂原激活的蛋白激酶依赖的信号通路的某个上游步骤而增强NGF诱导PC12细胞延伸大量突起。
Objective: To investigate the effects of total alkaloids from Codonopsis pilosula on nerve growth factor-induced differentiation of rat pheochromocytoma PC12 cells. METHODS: PC12 cells were cultured in the presence or absence of nerve growth factor (NGF) using total alkaloids of Codonopsis pilosula, and the extension of PC12 cells and related protein kinases were analyzed. RESULTS: Codonopsis alkaloids alone could not induce elongation of PC12 cells, but in the presence of very low concentrations of NGF (2 μg/L), total alkaloids in Codonopsis pilosula significantly induced a large number of protuberances in PC12 cells and enhanced mitogen-activated proteins. Phosphorylation of kinase (MAPK), and this potentiation can be inhibited by mitogen-activated protein kinase kinase inhibitor PD98059. CONCLUSION: Codonopsis total alkaloids enhance NGF-induced PC12 cell expansion by amplifying a certain upstream step of the mitogen-activated protein kinase-dependent signaling pathway.
