目的探讨新西兰白兔脱细胞真皮基质对人脂肪间充质干细胞(h Ad MSC)生长的影响。方法建立新西兰白兔脱细胞真皮基质,设立对照组、基质培养组。利用MTT实验检测h Ad MSC的增殖情况,激光共聚焦显微镜观察h Ad MSC在细胞基质上的生长状态;ELISA实验检测各组细胞上清液中基质金属蛋白酶1(MMP-1)、基质金属蛋白酶抑制剂1(TIMP-1)蛋白含量。结果与对照组相比,基质培养组h Ad MSC的细胞增殖OD值升高比较明显(P<0.05);h Ad MSC在脱细胞真皮基质上的生长状态良好,基质培养组MMP-1、TIMP-1蛋白的含量明显增高(P<0.01)。结论脱细胞真皮基质有利于促进h Ad MSC的生长,对h Ad MSC组织工程技术修复组织损伤等具有重要意义。
Objective To investigate the effect of acellular dermal matrix of New Zealand white rabbits on the growth of human adipose-derived mesenchymal stem cells (h AdMSCs). Methods New Zealand white rabbits acellular dermal matrix was established, the control group and the matrix culture group were established. MTT assay was used to detect the proliferation of h Ad MSC. Laser scanning confocal microscopy was used to observe the growth of h Ad MSC in the cell matrix. ELISA was used to detect the expression of matrix metalloproteinase 1 (MMP-1), matrix metalloproteinase Inhibitor 1 (TIMP-1) protein content. Results Compared with the control group, the OD value of h Ad MSC cultured in matrix was significantly increased (P <0.05). The hAd MSC had a good growth on acellular dermal matrix. The MMP-1, TIMP -1 protein content was significantly increased (P <0.01). Conclusion The acellular dermal matrix is helpful to promote the growth of h Ad MSC and is of great significance for the tissue engineering of h Ad MSC tissue repair.
