目的:探讨苏木含药血清对人肺癌PG细胞株的增殖抑制作用及对Cyclin D1、CDK4蛋白表达的影响。方法:将肺癌PG细胞分为空白组、苏木组、苏木+顺铂组、顺铂组4组,四唑盐比色法(MTT)中各组根据血清浓度的不同分为20%、10%、5%、2.5%四组,观察各组PG细胞的增殖抑制作用;免疫荧光法中各组血清浓度为20%,采用激光共聚焦显微镜观察各组Cyclin D1、CDK4蛋白表达。结果:MTT法中10%、20%苏木组细胞的增殖抑制率明显,具有作用时间和剂量依赖性,但低于顺铂组和苏木+顺铂组,苏木+顺铂组PG细胞的增殖抑制率与顺铂组比较,差异无统计学意义(P>0.05)。免疫荧光结果显示:与空白组比较,苏木组CDK4、Cyclin D1蛋白减少(P<0.05),顺铂组和苏木+顺铂组CDK4、Cyclin D1蛋白的表达亦减少(P<0.01);且顺铂组和苏木+顺铂组CDK4、Cyclin D1蛋白的表达低于苏木组(P<0.05)。与顺铂组比较,苏木+顺铂组CDK4、Cyclin D1蛋白的表达差异无统计学意义(P>0.05)。结论:苏木含药血清通过下调Cyclin D1、CDK4蛋白表达抑制PG细胞增殖。
Objective: To investigate the inhibitory effect of hematoxylin and eosin serum on the proliferation of human lung cancer cell line PG and its effect on the expression of Cyclin D1 and CDK4. Methods: The lung cancer PG cells were divided into blank group, suma group, Sumu + cisplatin group, cisplatin group 4 group, tetrazolium salt colorimetry (MTT) in each group according to the serum concentration is divided into 20% 10%, 5% and 2.5% respectively. The proliferation inhibition of PG cells in each group was observed. The serum concentration of each group was 20% by immunofluorescence method. The expressions of Cyclin D1 and CDK4 protein in each group were observed by laser scanning confocal microscopy. Results: MTT assay showed that 10%, 20% Hematoxylin-Eosin significantly inhibited the proliferation of Hematoxylin-Eosin (HE) cells in a time-and dose-dependent manner, but were lower than that in cisplatin group and Hematoxylin + cisplatin group There was no significant difference between the two groups (P> 0.05). Immunofluorescence results showed that compared with the blank group, the protein expressions of CDK4 and Cyclin D1 decreased (P <0.05) in Hematoxylin - Eosin and the expression of CDK4 and Cyclin D1 in cisplatin group and Hematoxylin - Eosin + cisplatin group decreased (P <0.01); The expression of CDK4 and Cyclin D1 protein in cisplatin group and hematoxylin + cisplatin group was lower than that in hematoxylin and eosin group (P <0.05). Compared with cisplatin group, the expression of CDK4 and Cyclin D1 in Sumu + cisplatin group had no significant difference (P> 0.05). Conclusion: The serum containing Hematoxylin-Erioglitazone inhibits the proliferation of PG cells by down-regulating the expression of Cyclin D1 and CDK4.
