目的:通过研究芒果苷对MAPK信号通路的影响,探讨芒果苷抑制HBV在细胞内复制的可能机制。方法:体外培养2215细胞,采用Western blot检测芒果苷对2215细胞MAPK信号转导通路中ERK、JNK、p38蛋白含量及磷酸化水平的影响;采用报告基因实验检测芒果苷对2215细胞内转录因子AP-1活性的影响。结果:芒果苷对2215细胞MAPK信号转导通路中ERK、JNK及p38三种蛋白含量及磷酸化水平均无影响。但可以不同程度的抑制TGF-β诱导的2215细胞MAPK信号转导通路中JNK蛋白的活性,对p38和ERK的活性无明显抑制作用。同时芒果苷可抑制AP-1的转录活性。结论:芒果苷可抑制2215细胞MAPK信号转导通路中JNK蛋白及其下游转录因子AP-1的活性。提示此通路可能是芒果苷抑制HBV在肝细胞内复制的机制之一。
OBJECTIVE: To investigate the effect of mangiferin on MAPK signal pathway and to explore the possible mechanism by which mangiferin inhibits HBV replication in the cell. Methods: 2215 cells were cultured in vitro. The effects of mangiferin on the levels of ERK, JNK and p38 protein and the phosphorylation of MAPK signal transduction pathway in 2215 cells were detected by Western blot. The reporter gene assay was used to detect the effect of mangiferin on 2215 cell intracellular transcription factor AP -1 activity. Results: Mangiferin had no effect on the levels of phosphorylation of ERK, JNK and p38 in MAPK signal transduction pathway in 2215 cells. But could inhibit the activity of JNK protein in MAPK signal transduction pathway of 2215 cells induced by TGF-β to varying degrees, and had no obvious inhibitory effect on the activity of p38 and ERK. At the same time, mangiferin can inhibit the transcriptional activity of AP-1. Conclusion: Mangiferin can inhibit the activity of JNK protein and its downstream transcription factor AP-1 in MAPK signal transduction pathway of 2215 cells. Suggesting that this pathway may be one of the mechanisms by which mangiferin inhibits HBV replication in hepatocytes.
