目的研究西南獐牙菜(Swertia cincta Burk.)全株的化学成分及其抗前列腺增生活性,为开发利用提供药理学研究依据。方法采用正、反相硅胶柱、RP-HPLC、凝胶柱、薄层制备等色谱技术,通过波谱分析进行结构鉴定;采用皮下注射丙酸睾酮建立SD(Sprague-Dawley)大鼠前列腺增生模型,观察各组前列腺指数的变化。结果从西南獐牙菜全株中分离到11个化合物,经波谱数据分析分别鉴定为獐牙菜苦苷(1),(R)-(-)-龙胆苦苷(2),獐牙菜苷(3),6’-O-β-D-葡萄糖基獐牙菜苷(4),6’-O-β-D-葡萄糖基龙胆苦苷(5),马钱苷酸(6),齐墩果酸(7),2α,3β-dihydroxyolean-12-en-28-oic acid-28-O-β-D-glucopyranoside(8),β-胡萝卜苷(9),4-hydroxyl-3,5-dimethoxyl-6-O-β-D-glucosebenzene(10),异红草苷(11),其中獐牙菜苦苷能显著性降低前列腺指数。结论化合物5和8为首次从獐牙菜属植物中分离得到;其中化合物1具有一定的抗前列腺增生作用。
Aim To study the chemical constituents of Swertia cincta Burk. And its anti-prostatic hyperplasia activity, and to provide pharmacological basis for the development and utilization of Swertia cincta Burk. Methods The structures were elucidated by spectral analysis using positive and reversed phase silica gel column, RP-HPLC, gel column and preparative TLC. Sprague-Dawley rat model of benign prostatic hyperplasia was established by subcutaneous injection of testosterone propionate. The changes of prostate index in each group were observed. Results Eleven compounds were isolated from the whole plant of Swertia brasiliensis, and were identified as swertiarin (1), (R) - (-) - gentiopicroside (2) Glycosides (6), 6’-O-β-D-glucosylcertenin (4) , Oleanolic acid (7), 2α, 3β-dihydroxyolean-12-en-28-oic acid-28-O-β-D-glucopyranoside (8) , 5-dimethoxyl-6-O-β-D-glucosebenzene (10) and isoswellin (11). Swertia glycosides significantly reduced the prostate index. Conclusion Compounds 5 and 8 were isolated from Swertia for the first time. Among them, compound 1 has a certain anti-prostatic hyperplasia effect.
